Production of Recombinant Humanized Monoclonal Anti-Human Neutrophil Antigen (HNA) Antibodies With Potential Applicability as Standard Antibodies

Antibodies against human neutrophil antigens (HNAs) play a part in several conditions: neonatal alloimmune neutropenia, autoimmune neutropenia, and transfusion-related acute lung injury (TRALI). Accurate testing for these antibodies is therefore essential for diagnosis and prevention. Current tests rely on reference antisera taken from people. These are in short supply, vary in strength, can contain other antibodies that interfere with results, and are limited by ethical rules on human materials. This CME activity is based on a study in Transfusion that produced the first humanized monoclonal antibodies to the HNA-1 and HNA-2 antigens, made from existing mouse antibodies. Laboratories in an International Society of Blood Transfusion (ISBT) workshop then tested them. The activity covers why reliable reference reagents matter, how these antibodies were made and checked, and what they could mean for neutrophil antibody testing in laboratories worldwide.
